Modification of Methods for RNA Isolation with Guanidine Thiocyanate and Vacuum Transfer to Membranes Improve the Possibility to Analyze High Molecular Weight Transcripts from Animal Cells

 

Summary

 

    A modified method of RNA isolation in the presence of guanidine thiocyanate from the animal cells is suggested. The method permits the purification of high molecular weight samples from nucleus as well as from cytoplasm. The technique is also developed for (luick RNA transfer using vacuum blotting from agarose gels to adsorbing membranes after electrophoretic separation of RNA. The efficiency of combination of the above two methods to analyze the size of individual gene transcripts and their distribution between the nucleus and cytoplasm is shown by hybridization of RNA blots with radioactive DNA probes.
