Cloning and overexpression of the part of envelope protein El gene of classical swine fever virus (Strain Shi-Min) in Escherichia coli

A fragment ofEl geneofvirulent strain Shi-Minofclassicalswinefevervirus(CSFV) 775 bpinlength has been cloned using reverse transcriptase-polymerase chain reaction (RT-PCR). Primary sequence of the cloned fragment has been determined. This fragment has been fused to glutathione-S-transferase and expressed in E. coli. Molecular weight of the part of El protein was estimated to be 31 kD as expected. The recomb inant protein has been found mainly in insoluble fraction, its yield was up to 15% of the total cellular protein. Possible use of the protein is discussed.
