Стероид-рецепторные комплексы и механизм регуляции транскрипции эукариотического генома

Abstract

Исследовано взаимодействие стероид-рецепторных комплексов с нуклеиновыми кислотами и хроматином in vitro и in vivo. Показано, что высокоочищенный глюкокортикоид-рецепторный комплекс обладает выраженной избирательностью ассоциации с нуклеиновыми кислотами в зависимости от их состава и последовательности оснований. Установлено, что глюкокортикоидные рецепторы печени крыс связываются с повышенным сродством с отдельными сайтами гомологичной ДНК; эти сайты вовлекаются в состав транскрипционно активного хроматина при воздействии глюкокортикоидов на живые клетки. Изучение метилирования ДНК печени крыс и ее чувствительности к ДНКазе І в ходе гормональной индукции привело к заключению, что гормонреактивные локусы хроматина находятся в особой «потенциально активной» конформации. Таким образом, гормональная чувствительность генов зависит как от последовательности ДНК, так и от способа хроматиновой организации гена; при этом внутриклеточная РНК может конкурировать с ДНК за связывание рецепторов и тем самым смягчать силу гормонального воздействия на хроматин.
