Abstract

Отримання рекомбінантних білків у штамах-продуцентах, створених на базі Escherichia coli, потребує виділення цільо­вого продукту з бактеріальних клітин. Використання бак­теріофага лямбда в таких системах суперсинтезу є одним з шляхів розв'язання цієї задачі. В результаті лізису фагом клітин Е. coli, що несуть плазміду з цільовим геном, рекомбінантний білок виходить у культуральне середовище. Од­нак в інфікованій клітині фаг може розвиватися як за літичним, так і лізогенним шляхом. Вибір літичного шляху роз­витку фага можуть обумовлювати різні фактори. Нами досліджено вплив температури культивування (37, 28 і 21 °С) інфікованих фагом X клітин Е. coli, що несуть плазміду з геном альфа-інтерферону людини (ІФН), на вихід розчинного ІФН у культу рольному середовищі. В цій роботі показано, що ефективність використання бактеріофага лямбда для лізису клітин продуцента і виходу розчинного ІФН у культуральне середовище залежить від температури культивування інфі­ кованих клітин Максимальна концентрація розчинного цільо­вого продукту в середовищі спостерігалася при температурі 21 °С, яка не є оптимальною для літичного розвитку фага. Обговорюються можливі причини отриманих результатів.
