Динаміка розщеплення поліедрів протеазою поліедрину вірусу ядерного поліедрозу тутового шовкопряду (Bombyx mori) та ідентифікація розщеплених зв'язків

Abstract

У процесі розчинення поліедрів вірусу ядерного поліедрозу (ВЯП) В. mori при рН 11 С і кімнатній температурі за 2 год протеаза поліедрів розщеплює в поліпептидному ланцюгу полі­ едрину (244 залишки амінокислот) пептидні зв'язки Tyr-lle (208–209), Tyr-Thr (186—187), Ala-Asn (121–122) та Trp-Ser (88–89). Жоден зв'язок не розщеплюється остаточно (100 %), згідно з даними електрофорезу в поліакриламідному гелі (ПААГ) з додаванням додецилсульфату натрію, що при­ зводить до появи набору поліпептидних ланцюгів, які перекри­ваються, з м. м. 28000 (цілий поліпептидний ланцюг), 25000, 22000, 14500, 10500 та 8000 (його фрагменти). Цим фрагмен­там поліпептид ного ланцюга відповідають (за даними первин­ної структури) такі фрагменти: 1–208 (м. м. за первинною структурою 24715), 1–186 (21980), 1–121 (14362), 1–88 (10443) і 122–186 (7633). Першим при 15-хв розчиненні розщеплюється зв'язок 208–209, за ним (45 хв) – зв'язок 186–187 і два останніх розщеплюються за 2 год. Із збіль­шенням часу розчинення результати не змінюються. Але при 2-год розчиненні за температури 37 °С додатково розщеплю­ються зв'язки Tyr-Leu (118–119), Phe-Val (193–194) та Tyr-Lys (203-204), причому зв'язки 203–204, 208–209, 88–89 та 118–119 розщеплюються повністю, а решта – частково. Це спричинює появу цілого набору фрагментів: 118–186, 122–186, 118–193, 122–193, 118–203, 122–203, 1–88 з м. м. 7633, 7916, 8445, 8728, 9765, 10048, 10443 відповідно, вирахуваними на основі первинної структури, що корелює з дифузною смугою з м. м. 7000–10000 в ПААГ. Дискутується вплив протеази поліедрів на такі процеси, як дисоціація– асоціація поліедрину в розчинах та асоціація фрагментів поліедрину з мембранами віріонів.
