Abstract

Методом диференційної УФ-спектроскопії вивчено вплив іонів Cd2+ на конформаційну рівновагу триланцюгового полінуклеотиду поліU·поліA·поліU (0,1 М Na+, pH 7). Показано, що ці іони не зв’язуються с гетероатомами азотистих основ поліA та поліU, що знаходяться у складі потрійної (A2U) та подвійної (AU) спіралей, та не змінюють їхньої конформації. Нагрівання A2U индукує два послідовних процеси: перший відповідає відділенню від A2U однієї нитки поліU, тобто реалізації переходу 3→2, другий – більш кооперативному плавленню AU, тобто переходу 2→1. Концентраційні залежності температур плавлення, які відповідають цим процесам, мають точку перетину (Tm=60 оС; [Cd2+] 3,5·10–4 M), при якій термостабільності A2U та AU стають однаковими. Теоретичними розрахунками концентраційних залежностей (Tm)3→2 показали, що результати досліду та теорії найкраще узгоджуються при ентальпії переходу ΔН3→2 = 5 ккалмоль–1триплет–1.
