Abstract

Вивчено комплексоутворення акридинового забарвника профлавіну з самокомплементарним дезокситетрарибонуклеозидтрифосфатом 5'-d(ApCpGpT) у водно-соляному розчині методом 1H-HMP-спектроскопії (500 МГц). Двомірна гомоядерна ПМР-спектроскопія (2M-COSY та 2M-NOESY) використана для повного віднесення сигналів протонів молекул в розчинах і для якісного аналізу характеру взаємодії профлавіна з тетрануклеотидом. Виміряно концентраційні залежності протонних хімічних зсувів молекул. На основі запропонованих моделей комплексоутворення визначені рівноважні константи реакцій та граничні значення хімічних зсувів протонів забарвника у складі комплексу. Досліджено відносний зміст комплексів різних типів в розчині та виявлені особливості динамічної рівноваги в залежності від співвідношення концентрацій забарвника і тетрануклеотида. Виходячи з приведених даних зроблено висновок про переважний зв'язок профлавіну з CG-сайтом тетрануклеотиду в мономірній та дуплексній формах. Побудована найбільш імовірна структура 1 : 2 комплексу забарвника з тетрануклеотидом, що базується на розрахункових значеннях індукованого хімічного зсуву протонів профлавіну і 2M-NOESY-спектpy.
