Зв'язування антибіотиків актиноцинового ряду з матрицею поліфосфату

Abstract

Методами УФ та Раманівської спектроскопії досліджено ком­плекси поліфосфату (ПФ) з аналогами актиноміцину D, по­ хідними актиноцину Actll і Actlll. Всі досліди виконували у фосфатному буферному розчині (рН 6,86), з сумарною концентрацією іонів Na+ і К+ , яка дорівнює 7×10–3 М Для розрахунків спектрів утворюваних комплексів та термодинамічних пара­метрів зв'язування лігандів з ПФ використано програму оптимізації спектрофотометричних концентраційних залеж­ностей DALSPH. Для двох типів лігандів методом спектро­фотометрії отримано значення констант і місць зв'язування, а також спектри агрегатів і мономерно зв'язаних похідних актиноцину. За допомогою Раманівської спектроскопії визна­чено основні групи атомів, які беруть участь в утворенні різних типів комплексів. Показано, що при мономерному зв'язуванні Actll та Actlll з ПФ, аміногрупа актиноцинового похідного зв'язується з групою Р-О матриці ПФ.
