Орієнтаційні ефекти, що викликаються присутністю в плазміді pUC фрагментів rplJL-rpoBC-оперону Escherichia coli

Abstract

Сконструйовано низку рекомбінантних плазмід pUC, що містять фрагменти rplJL-rpoBC-оперону E. coli. Встановлено, що односпрямовані орієнтації у плазмідах pUC притаманні фрагменту EcoRI (1568 п. о.), що містить ген rplJL з промотором PL10, і BspRI-A-фрагменту зі структурною частиною гена rplJL. Орієнтація обох фрагментів у рекомбінантних плазмідах така, що напрямок транскрипції гена rplJL вбудованого фрагмента і lacZ'-вектора – протилежний.
